Syndecan-4 in Tumor Cell Motility

چکیده
Syndecan-4 (SDC4) is a ubiquitously expressed, transmembrane proteoglycan bearing heparan sulfate chains. SDC4 involved in numerous inside-out and outside-in signaling processes, such as binding sequestration of growth factors extracellular matrix components, regulation the activity small GTPase Rac1, protein kinase C-alpha, level intracellular calcium, or phosphorylation focal adhesion kinase. The ability this to link actin cytoskeleton enables contribute biological functions like cell migration, proliferation, cytokinesis, cellular polarity, mechanotransduction. multiple roles tumor pathogenesis progression has already been demonstrated; therefore, expression was investigated several types. influences by regulating proliferation well migration affecting cell-matrix pathways. Here, we summarize general role motility.

Specific Syndecan-1 Domains Regulate Mesenchymal Tumor Cell Adhesion, Motility and Migration
BACKGROUND Syndecans are proteoglycans whose core proteins have a short cytoplasmic domain, a transmembrane domain and a large N-terminal extracellular domain possessing glycosaminoglycan chains. Syndecans are involved in many important cellular processes. Our recent publications have demonstrated that syndecan-1 translocates into the nucleus and hampers tumor cell proliferation. In the present...
متن کاملSyndecan-4 and integrins: combinatorial signaling in cell adhesion.
It is now becoming clear that additional transmembrane components can modify integrin-mediated adhesion. Syndecan-4 is a transmembrane heparan sulfate proteoglycan whose external glycosaminoglycan chains can bind extracellular matrix ligands and whose core protein cytoplasmic domain can signal during adhesion. Two papers in this issue of JCS demonstrate, through transfection studies, that synde...

Heparan sulfate proteoglycans are found on the surface of most cells. Syndecan-4 is a widely expressed transmembrane heparan sulfate proteoglycan. Using quantitative RNase protection assays and immunoblotting, syndecan-4 expression was characterized in 3T3-F442A mouse adipoblasts. These cells exhibit dramatic changes in their biological and morphological characteristics during differentiation t...

Tenascin-C is an adhesion-modulatory extracellular matrix molecule that is highly expressed in tumors. To investigate the effect of tenascin-C on tumor cells, we analyzed its antiadhesive nature and effect on tumor cell proliferation in a fibronectin context. Glioblastoma and breast carcinoma cell adhesion was compromised by a mixed fibronectin/tenascin-C substratum, which concomitantly caused ...
